I've installed USB 3 host adapters in many of my PCs that only had USB 2 ports.  If your machine has USB 3 ports there's no need.

I leave my USB 3 drives powered up 24/7.  When I boot my CPU the drives mount automatically.

I've tested raw SATA drives with 3 different interfaces:

1) hardwired to motherboard SATA ports
2) via USB 3
3) via FireWire 800

In my limited testing, primarily copying large files, neither the SATA nor the USB 3 connections limit the raw throughput of the drive mechanisms.  The FW800 connection is slower than the drives, though for most editing of DNxHD video it's perfectly usable.

Sure, a pair of internal drives in RAID 0 would be faster than an individual drive via USB 3.  But I'd guess that if you pull one of those internal 2TB drives and put it in an external USB 3 dock or case, its speed will be indistinguishable from the drive that remains inside.  

I haven't yet heard any L'ers report that a USB 3 connection has actually throttled the throughput of an Avid media drive, compared to any other connection.

But...
* I can not boot with all the USB 3.0 drives connected (so I first boot,
and then switch on the power of the external drives)
* the USB 3.0 drives fall asleep after a few minutes, if they are not in
use all the time - which is annoing.
(now I use KeepAliveHD http://tinyurl.com/lw2cwlq which works fine for me)
* of course the USB 3.0 drives are a little bit slower than the internal
raid 0 (2x2tb)

All USB 3.0 drives are connected directly to the USB 3.0 ports of the
comuter.
