I can't quite make out what you want to do; your use of
"multipathed" may be throwing me.
Each MacPro has two ethernet ports.
You can
1) Run any ethernet cable from a port on Mac #1 to Mac #2 to
network them. You will have to manually set compatible IP and
subnet so they can talk to each other. You end up with a single 1
gigabit network between the two Macs.
2) Run two ethernet cables from each port on Mac #1 to Mac #2,
and create two networks between them. You will have to manually
set the IP/subnets of the ports on each end of one cable to be
compatible. So, say, you end up with port 1 on Mac #1 with IP
192.168.0.1 and port 1 on Mac #2 with IP 192.168.0.2; an ethernet
cable connecting the ports. Then, port 2 on Mac #1 could be IP
10.0.0.1, port 2 on Mac #2 with IP 10.0.0.2; another ethernet
cable connecting those two ports.
Now you'll have two networks between the Macs running at 1
gigabit each. Share/mount drives on each of those networks and
copy away with greater throughput than just using one cable
between the Macs. Of course the assumes you have lots of stuff to
copy -- some over one network, some over the other. You can't
copy one big file over both networks at the same time. But total
date throughput will be close to double option 1) above (assuming
your drives can keep up.)
3) "Link Aggregation" (is this what you mean by "multipath"?)
lets you hook up those two ethernet ports on each Mac to the
other Mac, and run it as one network, giving you a single 2
gigabit network instead of the two 1 gigabit networks option 2)
above gives you. This article talks about how to set that up on
OSX server. You might be able to do it on the normal OS... can't
check for you as I'm at home and our MacPros are at work...

I have a very simple setup for this project with two mac pro towers each with its own set of media on eSATA drives... I'd like to connect both machines via both gigabit ethernet ports if possible.

If you want to donate to Red Cross quake relief, you can do so through your cell phone. Text redcross to 90999 to make a $10 donation. It will be on your next cell bill.
