You mentioned "Mercury Transmit". Are your playback issues with the iOXT only in Premiere?
I had noticed playback issues in Premiere with my AJA KONA card...on tutorial material that was AVC encoded MP4 video. Upon further investigation, I discovered it was the audio that the Kona did not like. The audio was compressed AAC. Switching the audio output to the Mac instead of the Kona (leaving video still handled by the Kona), the playback issue went away.

For color accuracy, you need to avoid computer displays...especially connected directly via Thunderbolt. Avid doesn't send out an accurate for color grading signal that way. You need a thunderbolt IO device, like that IoXT you have. That might have been a bum unit, because I have one too, use it with my macbook pro, and the system is very speedy. I see no different with or without it, performance wise. There are also the Decklink Ultrastudio mini monitor...or AJA T-Tap... And then an HDTV, or broadcast monitor.
