Cache files and log files can eat up many gigs of space without even being noticed. I had a deal recently (Mac Snowleper) where a runaway log file had caused the boot drive to go from approx 60gb free to just a few hundred megs free basically overnight. Of course that propagated to the mirrors and backups and became a bit of a clusterfark to sort out.

I also had a folder syncing utility (mis)configured to where it was keeping its own "restore" cache going - that was an extra 20gb or so that I was able to clear without issue. Most of those things may be hidden from immediate view… I use this to make things visible:

http://code.google.com/p/hide-unhide/ (download link is at the bottom).

If you had the FC Suite on there, all the additional support and fancy preview goop for DVDSP, Motion, STP, etc can usually be moved or reinstalled to a different location without problem. That's a metric ass-ton of bits right there.

There are myriad cache cleaners on the market - I've used the stuff from http://www.titanium.free.fr/ with good success, but YMMV.

Of course scraping off all the lolcat videos will get you back some gigs as well.
